How to Start Last Dose Missions

First, make sure you have the Los Santos Drug Wars update installed and can access the Acid Lab content. You must complete all six First Dose missions before The Last Dose becomes available. Head to the liquor store in Sandy Shores and speak with Ron (marked with an R on your map) to wrap up the initial storyline.
After completing these requirements, Dax will contact you via phone call. This call triggers access to The Last Dose missions and marks the beginning of the second phase of the Drug Wars storyline.
Once you receive Dax's call, go to The Freakshop and look for the "D" marker on your map. This location serves as your starting point for all five Last Dose missions. Each mission unlocks after completing the previous one, so you'll progress through them sequentially.

Last Dose Missions Guide
The Last Dose consists of five missions that conclude the Los Santos Drug Wars storyline. Each mission has specific objectives to complete and offers rewards upon finishing. You'll need to tackle these missions in order, as each one unlocks after completing the previous mission.
The five Last Dose missions are:
This is an Intervention

1. This is an Intervention

The first mission drops you straight into chaos at The Freakshop. Labrat and Dax are arguing about Labrat's drug-induced condition when armed men dressed in white storm the location and kidnap Labrat during the commotion.
Your objective is straightforward: eliminate all the remaining kidnappers who stayed behind. These enemies will be positioned around the Freakshop area, so clear them out systematically. Watch for enemies taking cover behind vehicles and structures, and be prepared for a decent firefight. After eliminating all hostiles, return to Dax to complete the mission. This first mission rewards you with $100,000 upon completion.
2. Unusual Suspects

After Labrat's kidnapping, Dax sends you on a detective mission to track down leads. He suspects either The Lost MC or the Hippies are behind the attack as payback from the First Dose events.
You'll hit two locations first: a Hippie camp and a Lost MC hideout. At each spot, eliminate all enemies except the leaders, then interrogate them for information. Both leaders claim they know nothing about the kidnapping, leaving you empty-handed.
Dax then directs you to a third location where a higher-ranking Hippy Boss operates. Clear out his crew and interrogate the boss. During a speakerphone conversation with Dax, the boss reveals he's seen white-coated people setting up some kind of wellness operation at a warehouse on Elysian Island.
Head to Elysian Island and locate the warehouse. You'll spot a Granger 3600LX parked outside - hack the keypad to get inside. The building appears empty, but search thoroughly to find five clues that confirm this group works with FriedMind Pharmaceutical Corporation. Grab the access keycard you find during your search.
Armed gunmen will arrive as you're leaving, so be ready for another firefight before escaping the area. The mission concludes once you've eliminated the threats and made your exit. Complete the mission to earn another $100,000 reward.
3. FriedMind

With the access keycard from the previous mission, you'll infiltrate FriedMind HQ, located in the Eclipse Medical Tower. Fight your way through security to reach the top floor and search for clues about Labrat's location.
You'll find a clipboard containing a memo that reveals Labrat has been moved to the building's basement. Grab the keys from this area, then battle back downstairs, where you'll meet up with Luchadora on the ground level.
Head into the basement together to find Labrat tied to a chair, heavily guarded by armed personnel. You'll also spot Isiah Friedlander's Virtue parked in the corner. Clear out all the guards and free Labrat from his restraints.
Just as you're about to escape, Friedlander arrives with two additional gunmen. He explains his plan for a wonder-drug, boasting that he controls 80% of the world's supply of precursor chemicals. Friedlander reveals that Labrat's mind is too damaged to be useful, but you can serve as his replacement.
During the tense standoff, Luchadora prepares to attack Friedlander with a folding chair. He kills the lights in the confusion and escapes into the darkness. Hotwire the SUV and escape with Labrat and Luchadora. You'll face pursuit from enemies in Schafter LWBs and Granger 3600LXs during your getaway to The Freakshop. Complete this mission to earn another $100,000.
4. Checking In

Dax discovers that Friedlander is testing his new drug at Parsons Rehabilitation Center in Richman Glen. He and Luchadora hop into his Journey II and ask you to follow them to the location. Dax mentions that some Fooliganz members are already scouting the area.
You can arrive before Dax if you want. When you get there, you'll find two Fooliganz with a Glendale Custom engaged in a firefight against Friedlander's men. Help eliminate all the hostile forces in the area.
Once the enemies are cleared, attempt to break through the front door. The door resists your efforts initially, then suddenly opens with a small explosion that releases a gaseous cloud around you and your teammates.
The drug takes effect immediately, sending you on a psychedelic trip. Friedlander's voice echoes throughout the experience as you battle through four waves of enemies. The final wave features the most challenging opponents yet - clones of yourself.
The trip reaches its climax when you must surrender and allow your own ego clones to "kill" you. Once you give in to this bizarre scenario, the drug-induced experience ends. This surreal mission rewards you with another $100,000 upon completion.
5. BDKD

The final mission puts you in the pilot's seat of a Velum 5-Seater that Luchadora has stolen and parked in the LS Storm Drain outside The Freakshop. Fly to LSIA, where Friedlander is attempting to escape to South America in a Cargo Plane.
Luchadora explains the plan: perform an airborne hijacking since there's too much product to handle on the ground. Fly close to the Cargo Plane and use the RubberNeck signal box on your dashboard to hack open the rear cargo door. Once it's open, fly the Velum directly into the cargo bay.
Fight through Friedlander's goons as you make your way to the cockpit. Just as you're about to take control of the plane, Friedlander emerges from a packing crate wearing a parachute and jumps out, escaping once again.
Fly the hijacked Cargo Plane to Sandy Shores Airfield, where Dax arrives with a black Mule truck. Start unloading the stolen product into the truck while police sirens approach in the distance. Defend Dax and the truck alongside Luchadora and other Fooliganz members as he continues loading.
Once the truck is full, you and Dax head back to The Freakshop while Luchadora stays behind to distract the police. Your job is to get the Mule truck to The Freakshop before the cops destroy it. Complete this final mission to earn $100,000 and conclude the Last Dose storyline.
Final Words
The Last Dose missions conclude the Los Santos Drug Wars storyline with five missions that each reward $100,000, totaling $500,000. The unique drug-induced sequences and airborne hijacking make these missions stand out from standard GTA Online content.
Stock up on armor, health items, and ammunition before starting. The missions unlock sequentially, so complete them in order to progress through the storyline and claim all rewards.
